objective: acetaminophen (apap) overdose causes acute liver injuries. studies show that stem cell factor (scf) and its receptor, c-kit, enhance liver recovery from apap-induced injuries in mice. in this study we explore the effect of scf on activity of glutathione s-transferase (gsts) enzymes which are considered to be important in apap metabolism. methods: we divided 45 balb/c mice into three groups. within each group there were three sub-groups of five mice per subgroup. the groups included: 1. apap (300 mg/kg b.w., i.p.); 2. scf (40 µg/kg b.w., i.p.) given.30 minutes after apap (300 mg/kg b.w., i.p.), and 3.control mice treated with normal saline. the mice were sacrificed at 1, 12 and 24 hours, respectively. hepatotoxicity was evaluated in the 24 hour group by histopathology and assessment of biochemical serum markers (alt and ast). we assessed the levels of scf receptor (c-kit) protein and gst enzyme activities in the liver tissues.  results: hepatotoxicity was induced by apap (300 mg/kg, b.w) as evident by both histopathological observations and a significant (p

Effect of Viola Extract on Acetaminophen-induced Hepatotoxicity in Mice

Introduction: Acetaminophen is a common analgesic that is toxic to the liver in the case of high dose and antioxidants such as herbs can partially inhibit the harmful effects of high dose of such substances. Materials and Methods: In this study, 42 mice weighing 25-30 g were randomly divided into six groups (n=7). The first group as the control group received 1 ml/kg (0.5 ml/kg 12 hours apart)...

Increased resistance to acetaminophen hepatotoxicity in mice lacking glutathione S-transferase Pi.

Overdose of acetaminophen, a widely used analgesic drug, can result in severe hepatotoxicity and is often fatal. This toxic reaction is associated with metabolic activation by the P450 system to form a quinoneimine metabolite, N-acetyl-p-benzoquinoneimine (NAPQI), which covalently binds to proteins and other macromolecules to cause cellular damage. At low doses, NAPQI is efficiently detoxified,...

Hepatoprotective activity of phloretin and hydroxychalcones against Acetaminophen Induced hepatotoxicity in mice

Polyphenolics form a major part of the dietary antioxidant capacity of fruits and vegetables have been identified as chemopreventive or anticancer agents. Hydroxychalcones are polyphenols abundantly distributed throughout the plant kingdom and are compounds with two aromatic rings (benzene or phenol) and an unsaturated side chain. In the present study, effect of phloretin (apple major fl...

Evaluation the protective effects of doxycycline on acetaminophen-induced hepatotoxicity in mice

Acetaminophen (APAP) toxicity threatens human health due to increased mortality associated with its overdose. Doxycycline (DC) because of its properties such as antioxidant and anti-inflammatory can be a good therapeutic strategy to treat the acute toxicity induced by APAP. Male mice were divided to six groups in two periods of 3 and 24-h as normal saline, APAP 400 mg/kg, DC 100 mg/kg and group...

Expression of cytochrome P450 and glutathione S-transferase in human bone marrow mesenchymal stem cells

Currently several studies are being carried out on various properties of mesenchymal stem cells (MSCs)however there are a few investigations about drug metabolizing properties of these cells. The aim of thisstudy was to measure the key factors involved in drug metabolism in human bone marrow MSCs. For thispurpose, cellular glutathione (GSH), glutathione Stransferase (GSTs) and...
